AB  - Pseudomonas aeruginosa is a Gram-negative
pathogen responsible for frequent hospital-acquired
infections of the bloodstream, the respiratory
tract, and the urinary tract. Quorum
quenching enzymes are recognized as an alternative
antivirulence approach targeting pathogenic
bacteria. The efficacy of YtnP lactonase in
reducing the virulence of P. aeruginosa MMA83
in vivo using Caenorhabditis elegans as a model
system was investigated. The recombinant YtnP
lactonase exhibits no cytotoxicity, demonstrated
by its lack of harmful effects on both the
immortalized human HaCaT cell line and two
strains of C. elegans (AU37 and N2 wild-type). In
a toxin-mediated killing liquid assay, the survival
rates of C. elegans AU37 mutant and N2 wildtype
strains infected with the clinical isolate P.
aeruginosa MMA83 significantly increased when
pre-treated with YtnP lactonase, compared to
untreated controls. Considering that virulence
factors expression is regulated by quorum sensing
(QS) signaling it is hypothesized that YtnP
lactonase prolongs the life span of C. elegans
by downregulating the QS and expression of
virulence factors of MMA83. The protective effects
of YtnP lactonase against MMA83-induced
pathogenicity in C. elegans, coupled with its absence
of cytotoxicity, position YtnP lactonase as
a promising prophylactic agent with antivirulence
properties.

AB  - Antimicrobial resistance (AMR) arises when
bacteria and other microbes stop responding
to medications. AMR is now recognized as one
of serious global health threats, repeatedly
appearing in the World Health Organization’s
(WHO) lists of urgent global health challenges,
including the 2024 list. It is taking a fatal toll
– nearly 5 million deaths globally per year are
associated with AMR, encompassing 1.27 million
directly attributed to AMR. The COVID-19
pandemic paved the way for aggravation of
bacterial AMR – primarily due to enhancement
in unspecific and unjustified prescription and
use of broad-spectrum antibiotics, resulting in
what is now recognized as „silent pandemic of
AMR“. Bacteriophages (phages) are natural and
specific predators of bacteria - viruses that can
infect, replicate inside and lyse arguably any
bacteria. Their therapeutic potential is being
hastily evaluated through different approaches:
in silico, in vitro, ex vivo and in vivo – in laboratory
animals as well as in human case and clinical
studies. Although the results are promising,bacteria rapidly develop resistance against
phages, which why the isolation and research
of new phages is needed. Our work is concentrated
on three bacterial species for which critical
priority by WHO has been declared – carbapenem-
resistant Acinetobacter baumannii,
Pseudomonas aeruginosa and Klebsiella pneumoniae.
Twenty distinct pathogenic strains of
A. baumannii, 6 K. pneumoniae and 6 P. aeruginosa
were used as targets for bacteriophage
isolation, and total of 14, 22 and 8 potentially
distinct phages were collected, respectively. All
strains were nosocomial isolates obtained from
various tissues, including from terminally ill patients.
Six phages were characterized in detail.
In particular, phage vB_AbaM_ISTD was applied
against A. baumannii in zebrafish embryo
model of systemic infection, and demonstrated
powerful therapeutic potential, eradicating the
infection. Interestingly, its DNA was characterized
with highly modified thymidine (amassing
1228 Da), making it the largest non-canonical
deoxynucleoside reported so far.

AB  - During the past decades several quorum sensing
inhibitors (QSI) of plant origin have been isolated
and chemically characterized. QSI agents
of plant origin represent potential alternative
or complementary approach to antibiotic treatment
of multidrug-resistant bacteria and infections
caused by bacterial biofilms. The aim of
the current study was to screen QSI activities
of horseradish root extracts obtained using
different organic solvents and different root
processing methods (drying at 40°C, 60°C or extraction
of fresh material). Common opportunistic
pathogen Pseudomonas aeruginosa MMA83
was used for QSI screen. RT-qPCR was used to
analyze the effect of the extract on the relative
mRNA levels of the genes QS (lasR, lasI, rhlR, rhlI,
mvfR, pqsH) and the genes involved in P. aeruginosa
MMA83 virulence (lasB, phzM, rhlC, algK,
pvdS). Chemical composition of extracts was
determined by UHPLC Q-ToF MS analysis. The
most active extract obtained using fresh roots
and hexane/ethyl acetate (1:1) solvent mixture
was able to significantly reduce content all examined
mRNA. Qualitative chemical analysis
reviled presence of 15 phenolic acids and their
derivatives, 9 flavonoids and 10 glucosinolates
in majority of examined extracts. It is significant
to emphasize that the most active QSI extract
did not contain a single one, out of ten dominant
glucosinolates, which have undergone to
hydrolysis yielding isothiocyanates and other
sulphur-containing compounds responsible for
QSI effects. Our results strongly indicate that
even mild thermal treatment (40°C) of horseradish
roots prior to extraction could lead to severe
reduction or loss of QSI activity.

AB  - Pseudomonas aeruginosa has been recognized as
a priority pathogen by World Health Organization,
due to the emergence of multidrug-resistant
(MDR) strains. Thus, new treatment options
such as antivirulence strategy is urgently needed.
This strategy is based on the disruption of quorum
sensing (QS) activity of this pathogen. The
focus of this research was to explore the anti-QS
activity of four selected medicinal mushrooms
(Lentinula edodes, Cantharellus cibarius, Trametes
versicolor and Pleurotus ostreatus) extracts on
MDR clinical isolate P. aeruginosa MMA83. Another
aim was to check their cytotoxicity on Caenorhabditis
elegans AU37 (glp-4(bn2) I; sec-1(km4).
Among three types of mushroom extracts - hot
water polysaccharide extracts (WPE), hot alkali
polysaccharide extracts (APE) and methanol
extracts (Met), APE extracts downregulated all
tested QS and virulence factors genes of P. aeruginosa
MMA83. The most prominent effect was
observed for C. cibarius APE extract, lowering
expression from 2-fold (for lasI gene) to 20-fold
for lasB gene. Extracts didn’t show cytotoxic effect
on C. elegans. The efficacy of APE extracts
in lowering the expression of QS and virulence
factors genes of P. aeruginosa MMA83 indicate

a priority pathogen by World Health Organization,
due to the emergence of multidrug-resistant
(MDR) strains. Thus, new treatment options
such as antivirulence strategy is urgently needed.
This strategy is based on the disruption of quorum
sensing (QS) activity of this pathogen. The
focus of this research was to explore the anti-QS
activity of four selected medicinal mushrooms
(Lentinula edodes, Cantharellus cibarius, Trametes
versicolor and Pleurotus ostreatus) extracts on
MDR clinical isolate P. aeruginosa MMA83. Another
aim was to check their cytotoxicity on Caenorhabditis
elegans AU37 (glp-4(bn2) I; sec-1(km4).
Among three types of mushroom extracts - hot
water polysaccharide extracts (WPE), hot alkali
polysaccharide extracts (APE) and methanol
extracts (Met), APE extracts downregulated all
tested QS and virulence factors genes of P. aeruginosa
MMA83. The most prominent effect was
observed for C. cibarius APE extract, lowering
expression from 2-fold (for lasI gene) to 20-fold
for lasB gene. Extracts didn’t show cytotoxic effect
on C. elegans. The efficacy of APE extracts
in lowering the expression of QS and virulence
factors genes of P. aeruginosa MMA83 indicate
that these extracts can reduce pathogenicity of
P. aeruginosa. Also, they possess one of the desirable

Short-term effect of Brevibacillus laterosporus supplemented diet on worker honey bee microbiome

AB  - Introduction: Brevibacillus laterosporus is a promising microbiological agent that can be used to prevent and control destructive diseases affecting honey bee colonies. In the presentstudy, the short-term
effect of the B. laterosporus BGSP11 bee diet on microbiota and mycobiota was investigated.
Methods: The honey bee diet was supplemented with spores of B. laterosporus BGSP11 at a concentration of 1×108 CFU/mL in sucrose solution. Metabarcoding analysis of the bee microbial community profile was performed based on 16S RNA (bacteriobiota) and Internally Transcribes Spacer (ITS) region
(mycobiota) obtained using MiSeq Illumina sequencing. The QIIME2 v2021.4 pipeline was used to analyze the obtained amplicon data library.
Results: The results show that the BGSP11 bee diet slightly altered the bee microbiota and did not lead
to potentially harmful changes in the bacterial microbiota. Moreover, it can potentially induce positive
changes, mainly reflected in the reduction of opportunistic bacteria. On the other hand, the treatment
had a greater effect on mycobiota. However, the changesin the bee mycobiome caused by the treatment
cannot be considered a priori as beneficial or harmful,since the interaction between the bee and its mycobiome is not sufficiently studied. The observed positive changes in the bee mycobiome are mainly
reflected in the reduction of phytopathogenic fungi that may affect the organoleptic and techno-functional properties of honey.
Conclusion: This pilot study suggests that the introduction of BGSP11 in beekeeping practice as a biological agent could be considered due to no harmful effects observed on the microbiota of bees.

AB  - Background: This study was carried out for an understanding of the antioxidant mechanisms of field pea varieties under osmotic stress conditions caused by a lack of water. The activities of antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APx) and glutathione reductase (GR) were examined and analyzed. The gene expression levels of Cu/Zn SOD, cAPx and GR genes were also examined. Methods: Osmotic stress was stimulated using PEG 6000 with the osmotic potential of -0.1 MPa in 10 days old plant seedlings. The activity of the antioxidant enzymes was measured in the shoots and roots of pea seedlings. The gene expression levels of genes coding antioxidative enzymes were examined by the reverse transcription polymerase chain reaction (RT-PCR) technique. Arabidopsis 18S rRNA was used as endogenous control. Result: Osmotic stress changed the activities of antioxidant enzymes in the shoots and roots of pea seedlings. Varieties more tolerant to osmotic stress showed a significant increase in antioxidant activities in shoots and roots, while sensitive varieties showed a significant decrease. The results of the analysis of the expression of genes, coding for antioxidant enzymes, showed that the reaction of the tested cultivars to ROS was the result of increased expression of the tested genes in tolerant cultivars, i.e. decreased expression in sensitive cultivars.

AB  - Thermal springs represent a habitat with extreme conditions that harbor a unique
microbial community adapted to thrive in this environment. In addition to the geothermal
springs in Iceland, the thermal springs of Vranjska Banja are considered the hottest in
Europe with a water temperature of 63-95°C. Due to global warming and climate change,
there is a growing need for knowledge about the biodiversity of extreme natural habitats.
Besides the exceptional importance of studying extremophilic microorganisms, the
difficulty in their cultivation limits the expanding necessity of research in this field. This
study provides information about the microbial community structure and physicochemical
characteristics of the thermal spring of Vranjska Banja. To determine and monitor the
microbiota diversity of the Vranjska Banja hot spring, for the first time, comprehensive
culture-independent metagenomic analysis in parallel with a culture-dependent
approach was applied. The culture-independent composition of bacterial communities of
the thermal water was investigated using MiSeq-Illumina technology and analyzed by
the computing environment QIIME2 v2021. The applied cultivation approach resulted
in the isolation of 17 strains belonging to genera Bacillus, Anoxybacillus, Hydrogenophilus,

the thermal water was investigated using MiSeq-Illumina technology and analyzed by
the computing environment QIIME2 v2021. The applied cultivation approach resulted
in the isolation of 17 strains belonging to genera Bacillus, Anoxybacillus, Hydrogenophilus,

stress conditions caused by a lack of water. The activities of antioxidant enzymes such as superoxide dismutase (SOD), catalase
(CAT), ascorbate peroxidase (APx) and glutathione reductase (GR) were examined and analyzed. The gene expression levels of
Cu/Zn SOD, cAPx and GR genes were also examined.
Methods: Osmotic stress was stimulated using PEG 6000 with the osmotic potential of -0.1 MPa in 10 days old plant seedlings. The
activity of the antioxidant enzymes was measured in the shoots and roots of pea seedlings. The gene expression levels of genes
coding antioxidative enzymes were examined by the reverse transcription polymerase chain reaction (RT-PCR) technique. Arabidopsis
18S rRNA was used as endogenous control.
Result: Osmotic stress changed the activities of antioxidant enzymes in the shoots and roots of pea seedlings. Varieties more
tolerant to osmotic stress showed a significant increase in antioxidant activities in shoots and roots, while sensitive varieties showed
a significant decrease. The results of the analysis of the expression of genes, coding for antioxidant enzymes, showed that the
reaction of the tested cultivars to ROS was the result of increased expression of the tested genes in tolerant cultivars, i.e. decreased
expression in sensitive cultivars.

AB  - Klebsiella pneumoniae is a global health threat and bacteriophages are a potential solution in combating pandrug-resistant K. pneumoniae infections. Two lytic phages, LASTA and SJM3, active against several pandrug-resistant, nosocomial strains of K. pneumoniae were isolated and characterized. Their host range is narrow and latent period is particularly long; however, their lysogenic nature was refuted using both bioinformatic and experimental approaches. Genome sequence analysis clustered them with only two other phages into the new genus Lastavirus. Genomes of LASTA and SJM3 differ in only 13 base pairs, mainly located in tail fiber genes. Individual phages, as well as their cocktail, demonstrated significant bacterial reduction capacity in a time-dependent manner, yielding up to 4 log reduction against planktonic, and up to 2.59 log on biofilm-embedded, cells. Bacteria emerging from the contact with the phages developed resistance and achieved numbers comparable to the growth control after 24 h. The resistance to the phage seems to be of a transient nature and varies significantly between the two phages, as resistance to LASTA remained constant while resensitization to SJM3 was more prominent. Albeit with very few differences, SJM3 performed better than LASTA overall; however, more investigation is needed in order to consider them for therapeutic application.

AB  - The hot spring Vranjska Banja is the hottest spring on the Balkan Peninsula with a water temperature of 63–95 °C and a pH value of 7.1, in situ. According to the physicochemical analysis, Vranjska Banja hot spring belongs to the bicarbonated and sulfated hyperthermal waters. The structures of microbial community of this geothermal spring are still largely unexplored. In order to determine and monitor the diversity of microbiota of the Vranjska Banja hot spring, a comprehensive culture-independent metagenomic analysis was conducted in parallel with a culture-dependent approach for the first time. Microbial profiling using amplicon sequencing analysis revealed the presence of phylogenetically novel taxa, ranging from species to phyla. Cultivation-based methods resulted in the isolation of 17 strains belonging to the genera Anoxybacillus, Bacillus, Geobacillus, and Hydrogenophillus. Whole-genome sequencing of five representative strains was then performed. The genomic characterization and OrthoANI analysis revealed that the Vranjska Banja hot spring harbors phylogenetically novel species of the genus Anoxybacillus, proving its uniqueness. Moreover, these isolates contain stress response genes that enable them to survive in the harsh conditions of the hot springs. The results of the in silico analysis show that most of the sequenced strains have the potential to produce thermostable enzymes (proteases, lipases, amylases, phytase, chitinase, and glucanase) and various antimicrobial molecules that can be of great importance for industrial, agricultural, and biotechnological applications. Finally, this study provides a basis for further research and understanding of the metabolic potential of these microorganisms.

Highly stable and versatile α-amylase from Anoxybacillus vranjensis ST4 suitable for various applications

AB  - α-Amylase from the thermophilic bacterial strain Anoxybacillus vranjensis ST4 (AVA) was cloned into the pMALc5HisEk expression vector and successfully expressed and purified from the Escherichia coli ER2523 host strain. AVA belongs to the GH13_5 subfamily of glycoside hydrolases and has 7 conserved sequence regions (CSRs) distributed in three distinct domains (A, B, C). In addition, there is a starch binding domain (SBD) from the CBM20 family of carbohydrate binding modules (CBMs). AVA is a monomer of 66 kDa that achieves maximum activity at 60–80 °C and is active and stable over a wide pH range (4.0–9.0). AVA retained 50 % of its activity after 31 h of incubation at 60 °C and was resistant to a large number of denaturing agents. It hydrolyzed starch granules very efficiently, releasing maltose, maltotriose and maltopentaose as the main products. The hydrolysis rates of raw corn, wheat, horseradish, and potato starch, at a concentration of 10 %, were 87.8, 85.9, 93.0, and 58 %, respectively, at pH 8.5 over a 3 h period. This study showed that the high level of expression as well as the properties of this highly stable and versatile enzyme show all the prerequisites for successful application in industry.

AB  - In the current study, honey bees’ diet was supplemented with spores of Brevibacillus laterosporus BGSP11 at concentration of 108 CFU/ml in sucrose solution and its short-term effects on their micro- and mycobiota have been analyzed using Illumina MiSeq sequencing. Obtained results indicate that this treatment does not lead to potentially harmful changes in the bacterial microbiome of worker bees, slightly affecting the composition of core microbiota. Moreover, several potentially beneficial changes have been observed. The treatment has led to a significant increase in the abundance of Snodgrassella alvi, and species from Lactobacillus and Bifidobacterium genera which play important roles in protection against several honey bee pathogens. Simultaneously, B. laterosporus enriched diet have led to almost complete eradication of Enterobacteriaceae family, the taxon that contains several putative pathogen species. On the other hand, the treatment affected mycobiota more profoundly, which was expected considering the greater instability compared to microbiota. Although the observed changes in honey bee mycobiome cannot be considered a priori beneficial or harmful, since the interaction between the bee and its mycobiome has not been sufficiently studied, certain beneficial consequences of the treatment have been observed. They are primarily reflected in the reduction of phytopathogenic fungi that can affect the organoleptic and techno-functional characteristics of honey. In addition, before introducing B. laterosporus in beekeeping practice as a biological agent for pathogen control it is necessary to perform more thorough studies of the impact on the honey bee microbiome, immune system, physiology and economic characteristics of honey bee colonies.

AB  - The aim of this study was to investigate the effects of chestnut tannins in metabolizable protein deficiency, on dry matter (DM) intake, growth performance of finishing lambs, feed efficiency and digestibility of nutrients. The study included 30 crossbred lambs, blocked by body weight (BW) and divided into three groups, depending on the concentrations of added chestnut tannins in the rations (0, 10 and 20 g/kg DM for CON, 10T and 20T group, respectively). Rations were determined by BW and expected average daily gain (ADG), with a deficiency in metabolizable protein of about 15%. All groups received the same amount of daily feed. Dry matter intake, ADG and indicators of efficiency such as the Kleiber ratio (KR) and protein efficiency ratio (PER) were estimated. The digestibility of the rations was determined in three collection periods. Dry matter intake did not differ among groups. Growth performances for the whole experiment were significantly better for the 20T group than CON (p  lt  0.05), while no differences were established for group 10T. Group 20T achieved the highest ADG and total gain (228 g/day +/- 27.8; 13.7 kg +/- 1.7), with the best average conversion of dry matter (4.4 kg DM/kg BW), KR (15.6), and PER (1.6). In all three collections, dry matter and crude protein digestibility was higher for CON, while the digestibility of ether extract was higher for the 20T group than CON. Based on these results, it can be concluded that a ration of chestnut tannins in the concentration of 20 g/kg DM, in conditions of protein deficiency, can have a positive effect on lamb performances. This can be partially explained by the ability of tannins to form complexes with proteins, which can change intestinal protein flow and utilization.
AB  - Das Ziel dieser Studie war, die Wirkungen von Kastanien-Tanninen in Rationen mit
unzureichender Versorgung mit metabolisierbarem Protein auf die Aufnahme von
Trockenmasse (DM), die Wachstumsleistung von Mastlämmern, den Futteraufwand und die Verdaulichkeit von Nährstoffen zu untersuchen. Die Studie umfasste 30 Kreuzungslämmer,
die durch das Körpergewicht (BW) und die Konzentrationen von zugesetzten
Kastanien-Tanninen in der Nahrung (0, 10 und 20 g/kg DM für die Kontrolle, 10T- bzw.
20T-Gruppe) in drei Gruppen unterteilt wurden. Die Rationen wurden durch das Körpergewicht
(BW) und die erwartete durchschnittliche tägliche Zunahme (ADG) mit einem
Mangel an metabolisierbarem Protein von etwa 15% definiert. Alle Gruppen erhielten

einer Konzentration von 20 g/kg DM Futter bei Proteinmangel einen positiven Effekt auf

von Tanninen erklärt werden, Komplexe mit Proteinen zu bilden, die den Proteinfluss

In the recent years, considerable attention has been given to selenium status since its deficiency is linked with various disorders and affects at least 13 % of world population. Additionally, mushrooms are known to possess pronounced capacity for absorption of various micronutrients, including Se, from soil/substrate. Here, we investigate the possibility of using Se-rich zeolitic tuff as a supplement for production of selenized mushroom. Furthermore, the impact of the enrichment on the activity of antioxidant enzymes and biological potential of Coriolus versicolor medicinal mushroom is studied. Experimental approach. Se(IV)- and Se(VI)-modified natural zeolitic tuff from the Serbian deposit Zlatokop was used as supplement for mushroom cultivation. To examine the effectiveness of selenium enrichment, we determined total selenium with inductively coupled plasma mass spectrometry (ICP-MS), together with the activity of antioxidant enzymes in fresh fruiting bodies and biological potential of methanolic extracts. Antioxidant activity was evaluated using the appropriate tests for: inhibition of lipid peroxidation, DPPH free radical scavenging assay, Fe(III)-reducing antioxidant power assay and ability of chelating Fe2+ ions. The antibacterial activity against foodborne pathogens was measured by broth microdilution assay. Additionally, chemical composition of the prepared extracts was studied using UV-Vis and Fourier transform infrared (FTIR) spectroscopy. Results and conclusions. Content of selenium detected in biofortified C. versicolor was even 470 times higher than in control on dry mass basis ((140.7 +/- 3.8) vs (0.3 +/- 0.1) mu g/g), proving that Se-rich zeolitic tuff is an excellent supplement for mushroom production. Furthermore, the results of monitoring the activity of antioxidant enzymes revealed that most of the Se-enriched mushrooms exhibited higher superoxide dismutase (SOD) and cata lase (CAT) and lower glutathione peroxidase (GSH-Px) activities than control. Due to higher amounts of enzymes, which can quickly catalyze the reduction of superoxide radicals, the quality of selenium-enriched mushrooms is preserved for a longer period of time. Investigation of biological potential indicated that Se-enriched mushroom methanolic extracts, generally, expressed enhanced antioxidant properties. Additionally, extracts showed antibacterial activity against all tested pathogenic microorganisms. Novelty and scientific contribution. Cultivation of mushrooms on Se-enriched zeolitic tuff is a new technological approach for obtaining Se-fortified food/supplements with enhanced antioxidant and antibacterial activities.

AB  - This study deals with the evaluation of the bioaccessibility and antioxidant properties of phenolic compounds from heat-treated skim goat-milk powder fortified with grape-pomace-seed extract, after in vitro gastrointestinal digestion. Ultra-high performance liquid chromatography coupled to diode array detection and mass spectrometry (UHPLC-DAD MS/MS) analysis confirmed the abundant presence of phenolic acids and flavan-3-ols in the grape-pomace-seed extract (SE) and heat-treated skim goat-milk/seed-extract powder (TME). After in vitro digestion of TME powder and recovery of total quantified phenolics, flavan-3-ols and phenolic acids were 18.11%, 24.54%, and 1.17%, respectively. Low recovery of grape-pomace-seed phenolics indicated strong milk protein–phenolic interactions. Electrophoretic analysis of a soluble fraction of digested heat-treated skim goat milk (TM) and TME samples showed the absence of bands originating from milk proteins, indicating their hydrolysis during in vitro gastrointestinal digestion. The digested TME sample had better antioxidant properties in comparison to the digested TM sample (except for the ferrous ion-chelating capacity, FCC), due to the presence of bioaccessible phenolics. Taking into account the contribution of the digestive cocktail, digested TME sample had lower values of total phenolic content (TPC), in vitro phosphomolybdenum reducing capacity (TAC) and ferric reducing power (FRP), compared to the undigested TME sample. These results could be attributed to low recovery of phenolic compounds. TME powder could be a good carrier of phenolics to the colon; thus, TME powder could be a promising ingredient in the formulation of functional food.

AB  - The beneficial effect of moderate wine consumption is attributed to its micronutrients, especially polyphenols and largely depends on the digestion process. This work aimed to examine the influence of in vitro simulated digestion in the presence of complex food matrix on antioxidant and cytotoxic activity of red wine. The obtained results showed that total phenolic content of wine sample after in vitro digestion was higher compared to undigested wine, while the antioxidant activity of these samples was similar before and after digestion. Furthermore, it has been noticed that digested wine showed cytotoxic activity on SKBR3 breast adenocarcinoma cells near 20% after 72 h of treatment. This pioneering study that examined biological potential of in vitro digested wine in the presence of complex food matrix indicate that antioxidant and cytotoxic activity of red wine is preserved after digestion.

Total amino acid and protein, as well as taste stimulating and growth promoting amino acid amounts were analyzed in coated feed samples. Results indicated that coating with BGHN14 homogenate increased the amount of soluble proteins and free Glycine on feed surface, as well as the availability of protein-bound (hydroxy-)Proline. This lab-scale research provides the basis for use of lactobacilli as resource efficient source of soluble nutrients for animal feedstuffs.

Brevibacillus laterosporus supplementation diet modulates honey bee microbiome

AB  - Among them, bacterial and fungal pathogens Paenibacillus larvae, Melissococcus pluton, Ascosphera apis and
Nosema ceranae play a major impact on honey bees colonies. Thus, developing alternative prophylactic and
curative strategies are urgently needed. The use of probiotic bacteria in honey bee supplemental feeding is
therefore promising to treat or prevent diseases. Brevibacillus laterosporus, Gram-positive endospore forming
bacilli, is recognised as one of the promising antibacterial and antifungal agents producer.
The aim of this study was to examine the short-therm effects of B. laterosporus supplemented diet on worker
honey bee microbiome.
Dry spores of B. laterosporus strain BGSP11 have been administrated through a sugar syrup diet to ten
colonies and a representative specimen of worker honey bees was taken before the start of the treatment
and immediately after the syrup was consumed. The microbial diversity was assessed before and after the
treatment using Illumina MiSeq sequencing platforms (ID Genomics service, Seattle, WA, USA). 16s rRNA
sequencing for bacterial community profiling and fungal Internally Transcribes Spacer for mycological taxa
profiling were used. The next-generation microbiome bioinformatics platform QIIME2 v 2021.4 was used for
filtering and denoising obtained sequences, calculation of diversity metrics and taxonomy assignment. The
feature classifier was trained using the Greengenes v 13_8 for bacterial taxa and fungal UNITE database v 8.3.
The results obtained in this study indicated statisticaly significant alfa diversity between control and experimental
group honey bee microbiota composition. The diversity abundance was higher in control comparing
to the group treated with B. laterosporus strain BGSP11 spores. There was no significant diference in Bray-
Curtis distance among two groups of analysed samples. Regarding to mycological abundance, composition
was completely different between two groups; control group had Claviceps as predominant genus, while in
treated group of honey bee microbiome Metschnikowia genus was prevalent, indicating that the presence of
fungal pathogens in treated group is highly diminished.
