Isolation and Characterisation of Bacteriocin and Aggregation-Promoting Factor Production in Lactococcus lactis ssp lactis BGBM50 Strain

AB  - Lactococcus locus ssp. lactis BGBM50, a producer of lactococcin G and aggregation-promoting factor, was isolated from selected lactic acid bacteria taken from semi-hard cheese traditionally produced in the village Zanjic, Montenegro. Strain BGBM50 harbours a number of plasmids of different sizes. Plasmid curing experiments showed that genes for bacteriocin production are located on pBM140, a plasmid 140 kb in length. PCR analysis with primers specific for lactococcin Q and G genes gave fragment of the expected size. In addition, after plasmid curing of strain BGBM50, different derivatives with altered phenotypes were obtained, among them BGBM50-34 strain, which retained bacteriocin synthesis but had enhanced aggregation ability.

Karakterizacija Klebsiella sp. izolovane iz izvorske oligomineralne vode za piće

AB  - Predmet biohemijske i molekularne karakterizacije bio je dominantan soj ST5 vrste Klebsiella sp. izolovan iz oligomineralne vode za piće kaptiranog izvora na Vlasini. Zbog podataka da pojedine vrste roda Klebsiella mogu imati medicinski značaj, kao oportuni patogeni, izvršili smo biohemijsku i molekularnu karakterizaciju soja ST5. Biohemijska karakterizacija soja urađena je pomoću manualnog identifikacionog sistema API 20E (bioMèrieux). Procena identifikacije dobijena je na osnovu baze podataka identifikacionog softvera (Apilabplus V 3.3.3 bioMèrieux). Molekularna karakterizacija urađena je PCR amplifikacijom 16S rDNK i sekvenciranjem metodom 'Thermal cycling sequencing'. Poređenje sekvence sa bankom podataka gena NCBI za 16S rRNK dobijena je najveća sličnost (95%) identičnosti ispitivanog soja sa sojem Klebsiella sp. označenim kao LB-2 (gi|110931878|gb|DQ831003.1) izolovanog u Kini. Biohemijska i molekularna karakterizacija dokazala je autohtoni karakter soja ST5 vrste Klebsiella sp.
AB  - The subject of this biochemical and molecular characterization was the dominant strain ST5 of Klebsiella sp. isolated from oligomineral drinking water from the capped spring in the Vlasina mountaineous region. Former data have proven that some types of the genus Klebsiella may be of medical importance as opportunistic pathogens which was the reason why the biochemical and molecular characterization of the strain ST5 had been done. The biochemical characterization of the ST5 strain was performed by means of the manual identification system API 20 E (bioMèrieux). The identification was based on the data base of the identification software (Apilabplus V 3.3.3 bioMèrieux). The molecular characterization was performed by means of the PCR amplification of 16S rDNK and the sequencing method 'Thermal cycling sequencing'. The comparison of the NCBI gene bank data for 16S rRNK resulted in the highest level of similarity, i.e. 95% of similarity for the observed genus compared to the genus Klebsiella sp. marked as LB-2 (gi|110931878|gb|DQ831003.1) isolated in China. The biochemical and molecular characterization proved the autochthonous character of the ST5 strain of Klebsiella sp.
