AB  - In last decades, bacterial resistance to first choice
antibiotics has been drastically increasing, therefore,
the research of new antimicrobial substances
is of great importance. This rising problem
with bacterial resistance to existing antibiotics
affects not only the health care institutes but also
food plants. S. aureus and S. enteritidis pose a
number of challenges to the food industry and
cause foodborne illness in humans. In addition,
due to their favourable elemental composition,
oyster mushrooms (Pleurotus sp.) are a reservoir
of bioactive compounds that give them remarkable
antibacterial potential. P. ostreatus is of
great economic importance and is the second
most cultivated edible mushroom, therefore information
about its possible targets on bacterial
cells is of great importance for use as a dietary
supplement or medicinal purposes. The results of
the antibacterial assay showed that tested bacterial
strains were susceptible to the methanol extract of P. ostreatus (PoME), while microbicidal
activity was only detected against Gram-positive
bacteria. Scanning electron microscopy (SEM)
micrographs suggested that extract acted on
cytoplasmic membrane of S. aureus, while the
cell envelope of S. Enteritidis was the most likely
target. Natural extracts may outperform individual
bioactive compounds due to the synergistic
interaction between the metabolites, which can
enhance the effects of the individual components.
Extracts rich in antibacterials are emerging
as alternatives to synthetic antibiotics in the food
and health sectors. Among these, crude mushroom
extracts are particularly sought after for
their diverse bioactive ingredients, as they can
combat resistant strains of bacteria due to their
different targets and modes of action. PoME can
be used as an effective antimicrobial agent, suitable
for applications aiming to eradicate foodborne
pathogens, thus enhancing food safety.

AB  - Carbon quantum dots as a novel type of carbon nanomaterials have attracted the attention of many researchers because of their
unique optical, antibacterial, and anticancer properties as well as their biocompatibility. In this study, for the first time, carbon
quantum dots were prepared from o-phenylenediamine dissolved in toluene by a solvothermal route. Subsequently, the prepared
carbon quantum dots were encapsulated into polyurethane films by a swelling–encapsulation–shrink method. Analyses of the
results obtained by different characterization methods (AFM, TEM, EDS, FTIR, photoluminescence, and EPR) indicate the significant influence of the precursor on structural, chemical, and optical properties. Antibacterial and cytotoxicity tests showed that these
dots did not have any antibacterial potential, because of the low extent of reactive oxygen species production, and showed low dark
cytotoxicity. By investigating the cellular uptake, it was established that these dots penetrated the HeLa cells and could be used as
probes for bioimaging.

The solvent-cast films presented more ordered structures, higher mechanical resistance, elongation, and water vapor barrier performance than the electrospun mats. The SE-incorporating formulations showed phenolics’ delivery ability to food simulants influenced by structure, SE content, and media polarity. The electrospun mats expressed higher DPPH• radicals’ inhibition, while the solvent-cast films showed stronger Staphylococcus aureus and Escherichia coli growth inhibition, increased by SE incorporation. All formulations showed rapid complete bio-disintegration in compost (18–25 days).

AB  - The quest for sustainable biomaterials with excellent biocompatibility and tailorable properties has put polyhydroxyalkanoates (PHAs) into the research spotlight. However, high production costs and the lack of bioactivity limit their market penetration. To address this, poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) was combined with a bacterial pigment with strong anticancer activity, prodigiosin (PG), to obtain functionally enhanced PHBV-based biomaterials. The samples were produced in the form of films 115.6–118.8 µm in thickness using the solvent casting method. The effects of PG incorporation on the physical properties (morphology, biopolymer crystallinity and thermal stability) and functionality of the obtained biomaterials were investigated. PG has acted as a nucleating agent, in turn affecting the degree of crystallinity, thermal stability and morphology of the films. All samples with PG had a more organized internal structure and higher melting and degradation temperatures. The calculated degree of crystallinity of the PHBV copolymer was 53%, while the PG1, PG3 and PG3 films had values of 64.0%, 63.9% and 69.2%, respectively. Cytotoxicity studies have shown the excellent anticancer activity of films against HCT116 (colon cancer) cells, thus advancing PHBV biomedical application potential.

The treatment of these diseases is of utmost interest to further prevent the development of multi-drug resistance in different pathogens. Carbon quantum dots, as a new member of the carbon nanomaterials family, can potentially be used as a highly promising visible-light-triggered antibacterial agent. In this work, the results of antibacterial and cytotoxic activities of gamma-ray-irradiated carbon quantum dots are presented. Carbon quantum dots (CQDs) were synthesized from citric acid by a pyrolysis procedure and irradiated by gamma rays at different doses (25, 50, 100 and 200 kGy). Structure, chemical composition and optical properties were investigated by atomic force microscopy, transmission electron microscopy, X-ray photoelectron spectroscopy, Fourier transform infrared spectroscopy, Raman spectroscopy, UV-Vis spectrometry and photoluminescence. Structural analysis showed that CQDs have a spherical-like shape and dose-dependent average diameters and heights. Antibacterial tests showed that all irradiated dots had antibacterial activity but CQDs irradiated with dose of 100 kGy had antibacterial activity against all seven pathogen-reference bacterial strains. Gamma-ray-modified CQDs did not show any cytotoxicity toward human fetal-originated MRC-5 cells. Moreover, fluorescence microscopy showed excellent cellular uptake of CQDs irradiated with doses of 25 and 200 kGy into MRC-5 cells.

AB  - Long-term frozen storage of caprine milk is an important way of overcoming seasonal production. White brined cheese, matured for 56 days, was made from frozen-stored caprine milk. Several properties of experimental and control cheese variants were investigated throughout the maturation period: physico-chemical, sensory, proteolytic (by urea-PAGE electrophoresis), texture attributes, microstructure (by scanning electron microscopy). Most of investigated properties were influenced by frozen storage and maturation period, but differences were less pronounced on the 56th day of maturation. Moreover, sensory analysis showed that both cheese variants were well accepted by consumers.

AB  - Due to their low cost and possible green synthesis, high stability and resistance to photobleaching, graphene quantum dots (GQDs) can be considered as one of the class of carbon nanomaterials which may have great potential as an agent for photosensitized oxygen activation. In such a way, GQDs can be used as a theranostic agent in photodynamic therapy. In this work pristine GQDs, GQDs irradiated with gamma rays and GQDs doped with N and N, S atoms are produced using a simple, green approach. By using different techniques (AFM, HRTEM, SEM-EDS, FTIR, XRD, PL and UV-Vis) we investigated structural and optical properties of the new types of GQDs. We showed that GQDs functionalized with thiourea (GQDs-TU) completely lost the ability to produce singlet oxygen (O-1(2)) upon photoexcitation while functionalization with urea (GQDs-U) improves the capability of GQDs to produce O-1(2) upon the same conditions. Thus, presented GQDs modification with urea seems like a promising approach for the production of the efficient photosensitizer. On the opposite, GQDs-TU are efficient . OH quencher. Due to high singlet oxygen production and low cytotoxicity below 100 mu g/mL against HeLa cells, GQDs-U is a good candidate as an agent in photodynamic therapy at this concentration.

AB  - The addition of heteroatoms to pristine carbon quantum dots (CQDs) change their structure and optical properties. In this study, fluorine (F)- and chlorine (Cl)-doped CQDs are prepared by the one-step green hydrothermal route from sodium fluoride, sodium chloride, urea, and citric acid as the starting precursors. Microscopy analysis reveals that the average size of these quantum dots is 5 +/- 2 nm, whereas the chemical study shows the existence of C-F and C-Cl bonds. The produced F- and Cl-doped CQDs have fluorescence quantum yields of 0.151 and 0.284, respectively, at an excitation wavelength of 450 nm. Charge transfer resistance of F- and Cl-doped CQDs films is 2 orders of magnitude higher than in the pristine CQD films. Transport band gap of the doped CQDs is 2 eV bigger than that of pristine CQDs. Radical scavenging activity shows very good antioxidant activity of doped CQDs. Antibacterial testing reveals poor antibacterial activity against Staphylococcus aureus and Escherichia coli. The F- and Cl-doped CQDs are successfully used as fluorescent probes for cell imaging as shown by confocal microscopy.

AB  - Bacterial nanocellulose (BNC) emerged as an attractive advanced biomaterial that provides desirable properties such as high strength, lightweight, tailorable surface chemistry, hydrophilicity, and biodegradability. BNC was successfully obtained from a wide range of carbon sources including sugars derived from grass biomass using Komagataeibacter medellinensis ID13488 strain with yields up to 6 g L-1 in static fermentation. Produced BNC was utilized in straightforward catalyst preparation as a solid support for two different transition metals, palladium and copper with metal loading of 20 and 3 wt%, respectively. Sustainable catalysts were applied in the synthesis of valuable fine chemicals, such as biphenyl-4-amine and 4'-fluorobiphenyl-4-amine, used in drug discovery, perfumes and dye industries with excellent product yields of up to 99%. Pd/BNC catalyst was reused 4 times and applied in two consecutive reactions, Suzuki-Miyaura cross-coupling reaction followed by hydrogenation of nitro to amino group while Cu/BNC catalyst was examined in Chan-Lam coupling reaction. Overall, the environmentally benign process of obtaining nanocellulose from biomass, followed by its utilisation as a solid support in metal-catalysed reactions and its recovery has been described. These findings reveal that BNC is a good support material, and it can be used as a support for different catalytic systems.

The mineral phase within the composite films was identified as hydrozincite, Zn-5(CO3)(2)(OH)(6). The presence of the mineral phase improved, both mechanical and water vapor permeability properties of the obtained composite films, in a concentration-dependent manner. The release of zinc ions from composite films is in accepted levels (up to 2.5%), and sufficient to provide complete inhibition growth of S. Aureus. The results of this study suggest that agar/Zn-carbonate composites could be potentially used as affordable, eco-friendly and functional materials with tunable properties for food packaging, agriculture or biomedical application. In situ procedure offers possibilities for tailoring the physical-chemical properties of composite films, by varying the Zn-mineral phase load.

AB  - Photoactive materials called photosensitizers can be used for treatment of different types of cancer in combination with light source. In this paper, we have investigated pro-oxidant and antioxidant potentials of four graphene based nanomaterials (graphene oxide-GO, graphene quantum dots-GQDs, carbon quantum dots-CQDs and N-doped carbon quantum dots-N-CQDs) depending on the presence/absence of visible light source. Structural and optical properties of these materials and their potentials for reactive oxygen species generation/quenching are investigated by applying different microscopy and spectroscopy techniques (transmission electron microscopy, FTIR, UV-Vis, photoluminescence, electron paramagnetic resonance). Results show that all types of quantum dots has pro-oxidant and antioxidant potentials whereas GO demonstrated only moderate antioxidant effect. The best free radical scavenger is CQDs sample in the absence of light. CQDs are the best singlet oxygen generator under blue light irradiation as well. To check photo-cytotoxicity of these materials, photo-cytotoxic concentrations of the GO, GQDs, CQDs and N-CQDs were determined for three cellular lines: human rhabdomyosarcoma (RD), cell line derived from human cervix carcinoma Hep2c (HeLa) and fibroblast cell line from murine (L2OB). Cytotoxicity test has indicated that all samples are much less photocytotoxic than cis-diamminedichloroplatinum (cis-DPP). The production method and doping of quantum dots affect the photodynamic activity of tested samples very much.

AB  - New agar-based composite films with increasing Cu-carbonate and Cu-phosphate mineral phase content were prepared by in situ mineralization and solvent casting method. SEM and optical analysis revealed that Cu-carbonate phase had better compatibility with agar matrix than Cu-phosphate phase. Incorporation of both mineral phases improved mechanical and water vapor barrier properties of the obtained mineralized films, in concentration dependent manner. When 5 mM of carbonate precursor was incorporated into agar matrix, mechanical resistance was enchanced for 44% and water vapor barrier property for 40%. The release of Cu (II) was higher in acidic conditions for both mineralized composites and remained in the range of specific release limits for this metal. In addition, both mineralized composite films exhibited distinctive antimicrobial activity against Gram-negative (Escherichia coli) and Gram-positive (Staphylococcus aureus) bacteria. Overall, the Cu-carbonate and Cu-phosphate mineralized agar films showed potential to be used for food packaging materials, agriculture or medical purposes.

AB  - New mineralized, agar-based nanocomposite films (Zn-carbonate and Zn-phosphate/agar) were produced by a combination of in situ precipitation and a casting method. The presence of minerals significantly influenced the morphology, properties and functionality of the obtained nanocomposites. Reinforcement with the Zn-mineral phase improved the mechanical properties of the carbonate-mineralized films, but had a negligible effect on the phosphate-mineralized samples. Both nanocomposites showed improved optical and thermal properties, better Zn(II) release potential in a slightly acidic environment and exhibited antimicrobial activity against S. aureus. These results suggest that Zn-mineralized agar nanocomposite films could be potentially used as affordable, eco-friendly and active food packaging materials.

AB  - Electrochemically exfoliated graphene is functionalized graphene with potential application in biomedicine. Two most relevant biological features of this material are its electrical conductivity and excellent water dispersibility. In this study we have tried to establish the correlation between graphene structure and its antibacterial properties. The exfoliation process was performed in a two electrode-highly oriented pyrolytic graphite electrochemical cell. Solution of ammonium persulfate was used as an electrolyte. Exfoliated graphene sheets were dispersed in aqueous media and characterized by atomic force microscopy, scanning electron microscopy, Raman spectroscopy, Fourier transform infrared spectroscopy, X photoelectron spectroscopy, X-ray diffraction, electron paramagnetic resonance, zeta potential, contact angle measurements and surface energy. Antibacterial assays have shown lack of the significant antibacterial activity. Major effect on bacteria was slight change of bacteria morphology. Membrane remained intact despite significant change of chemical content of membrane components.

AB  - Bioactive compounds obtained from Coriolus versicolor (Trametes versicolor (L: Fr) Lloyd, 1920.) mushrooms cultivated in a stirred-tank bioreactor were tested to determine their antimicrobial potential. Extracellular polysaccharides were isolated from the fermentation broth by ethanol precipitation. A methanol extract was prepared from mycelium. The cultivation conditions applied during the fermentation process provided for significant biomass 6.63 +/- 0.31 g dry weight L-1 and yield of extracellular polysaccharides (EPS) (0.74 +/- 0.12 g L-1). Microscopic analyses revealed that the mycelium grew predominately in the form of fluffy pellets. The methanol extract demonstrated very good activity against all the tested Gram-positive bacteria. Bacillus spizizeni and Staphylococcus epidermidis were the most sensitive strains (minimum inhibitory concentration (MIC)  lt 0.3125 mg mL(-1)). Among the Gram-negative bacteria, Yersinia enterocolitica had the lowest MIC value, 5 mg mL(-1). Microbicidal activity of mycelia methanol extract was established in seven out of ten tested Gram-negative bacteria strains with minimum bactericidal concentration (MBC) values ranged from 20 to 40 mg mL(-1). Enterococcus faecalis and Staphylococcus aureus showed higher sensitivity to the extracellular polysaccharides (MIC values 2.5 mg mL(-1)). FTIR analysis revealed a more complex chemical composition of the methanol extract compared to EPS, which might explain the better antibacterial activity of the methanol extract. Our results suggested that the submerged cultivation of Coriolus versicolor followed by ethanol precipitation of EPS and the methanol extraction of mycelia can be a promising process to obtain biological active compounds with significant antimicrobial activity. Industrial Relevance: Mushrooms contain a large number of chemicals with potential use as antimicrobial compounds. One of the biggest challenges for providing biologically active compounds from mushrooms is short-term process standardization with a low risk for contamination. Submerged culture cultivation is the best choice for providing antimicrobial compounds from mushrooms. The submerged culture method represents an effective and energy-efficient means to produce novel antibacterial compound from mushrooms. Antibacterial activity testing revealed that methanol extract and isolated exopolysaccharides exhibited strong antibacterial activity, especially against Gram-positive bacteria.

AB  - The antibacterial activity of methanol extract obtained from fruiting body of industrially grown basidiomycete Coriolus versicolor was examined. The Minimum Inhibitory Concentration (MIC) values against various bacteria ranged from 0.625 to 20 mg mL(-1). versicolor expressed bactericidal activity against both Gram-positive and Gram-negative bacteria. The growth curves of Staphylococcus aureus and Salmonella enterica serovar Enteritidis, measured at 630 nm, and confirmed with macrodilution method showed that the obtained extract could inhibit the growth of tested bacteria. Scanning electron microscopy (SEM), transmission electron microscopy (TEM), and the loss of 260-nm-absorbing material were used to examine the ultrastructural changes in bacteria induced by the extract. When S. aureus was exposed to the MIC of C. versicolor, elongated and malformed cells were observed by SEM, while S. Enteritidis treated cells appeared shorter and aggregated with ruptured cell walls. TEM revealed the formation of non-membrane-enclosed bodies and depleted inner content of S. aureus. Larger and irregular periplasmic space and deformed and scattered components of the cell envelope were observed in treated S. Enteritidis. The loss of 260-nm-absorbing material indicated that the disruptive action of the extract on cytoplasmic membrane was more pronounced in S. aureus than in S. Enteritidis treated cells. The UV and FTIR spectrophotometric analyses revealed diverse composition of C. versicolor extract and high content of total phenolics. Altogether, mushroom extracts could be used to develop nutraceuticals or drugs effective against pathogenic microorganisms.
